Abstract

Background: To some extent, the most apparent conception of infertility is the inability to procreate after a year or more of unprotected coitus.. There are several risk factors, the most important of them is obesity the chronic metabolic diseases that is major contributors to female infertility. The amount of adipose tissue (body fat) in your body directly correlates with the level of leptin (LEP)  in our blood, obesity results in high levels of leptin (hyperleptinemia). Since the level of leptin is directly related to the genetic variation (polymorphism) of the leptin genes; so the present findings were carried out to evaluate the association between infertility with obesity and leptin gene polymorphism at rs10244329 position.

Material and methods:  By Allele Specific -PCR primers technique all isolated genomic DNA samples were genotyped.

Results: Findings showed that the genotypes frequency for control group was      30(50%), 26(43.4%), and 4(6.6%) for AA, AT, and TT respectively, while the allelic frequencies were 86(71.4%) and 34(28.6) for A and T allele respectively. While for cases group was (80.0%), (16.6%), and (3.4%) for AA, AT, and TT respectively. On the other hand the statistical distribution of A and T alleles for leptin gene at rs10244329 position for both studded groups was (28.4 % and 11.6%) for control and cases groups respectively.

Conclusion: Results concluded that the A allele may play as risk factor in present study (OR > 1.0) while the T allele may it took on the role of protection factor ( OR < 1.0).
