Abstract

Chalcones and their derivatives are emerging as a class of drugs with promising potential for use in oncology, particularly in the field of oncology. Chalcones consist of a linear α- and variable α, β-unsaturated carbonyl backbone and exhibit diverse biological activities, including anti-inflammatory, antimicrobial, antioxidant, and anticancer properties. Their structural modifications allow for targeted modifications that enhance the leading and receptor efficacy. This review focuses on recent advances in the synthesis, structural modification, and anticancer properties of chalcones. Their mechanisms of action are well understood and include induction of apoptosis, cell cycle arrest, inhibition of angiogenesis, and regulation of key signaling pathways such as NF-κB, PI3K/Akt, and MAPK. In addition, examples of chalcone hybrids and their efficacy against various cancer cell lines are presented, demonstrating the importance of structural modifications in enhancing cytotoxicity and selectivity. Chalcones are also being investigated as potentially safe and effective alternatives to current chemotherapeutic drugs, with interesting applications in overcoming drug resistance. This review aims to provide insight into the design and development of chalcones for cancer treatment and to lay the foundation for future advances in medicinal chemistry.
