Abstract
Exposure to ethidium bromide (EtBr) may cause toxic effects on male reproductive system, while black seeds oil components exert beneficial effects on enhancing its functionality. A fabricated Nigella sativa oil-loaded nanocomposite was created to guarantee that the components reach the target tissues in high concentrations and are not lost or altered. The aim of this study is to examine the impact of Polycaprolactone/ Chitosan/Nigella Sativa Oil Nanofibrous (PCNSONF) on the male rats reproductive functions when exposed to ethidium bromide. For this purpose, six groups of male rats, 12 each, received, for 30 days, a daily dosage of distilled water (T1), EtBr (T2), PCNF (T3), PCNF and EtBR (T4), PCNSONF (T5), and PCNSONF and EtBR (T6). Each group was allocated to two subgroups, sacrificed after 15 or 30 days of treatments, respectively.
In both treatment periods, when compared to the control, treatment with EtBr resulted in a significant decrease in the genital organ’s weight, decreased serum levels of GnRH, FSH, LH, and testosterone, decreased expression levels of testicular LHR, FSHR, ABP, 3β-HSD, and 17β- HSD, and pituitary FSHβ and LHβ genes, and pronounced pathological changes of testes. In contrast, both PCNSONF-treated groups (with or without EtBr exposure) demonstrated efficacy in improving these metrics when compared to equivalent groups. It is concluded that PCNSONF not only shields the reproductive organs from EtBr exposure but also works well to improve the efficacy and quality of male reproduction.
